The Gerudo Sanctuary can only be accessed by collecting the seven marked orbs in The Mysterious Eighth quest to obtain the eighth, larger orb. This then needs to be taken to the entrance in the North Gerudo Ruins and placed in the hollow at the opening to the Sanctuary, raising the bars on the door.
