Trial of the Labyrinth is a Shrine Quest from Breath of the Wild.
Begin the Quest
As soon as Link enters the Lomei Labyrinth Island, Link will begin the shrine quest. Actually climbing up to the Labyrinth is a bit difficult, given the height of the walls. Link will need to either have a massive amount of stamina, or will need to use Revali's Gale, and it is recommended to have the Climbing Set to speed up the process. Alternatively, you can glide from the Akkala Ancient Tech Lab area.
Guide
The Labyrinth is a maze that Link will need to navigate to reach the shrine that is right at the center-north of Labyrinth. While there are a variety of treasure chests to collect, the pathway to the actual shrine is fairly simple. From the entrance of the maze, Link can run all the way forward and then left, entering the maze at the top-left portion of the open area. From there, Link can head north where there a metal block. After making a right turn, the path ends up at a ladder that Link can climb up. After climb up a series of staircases, Link will reach the shrine, completing the shrine quest. Optionally, if you have the Climbing Set, you can climb to the top of the maze and simply jump down to the entrance, bypassing the entire maze portion and dodging all of the guardians.
Tu Ka'loh Shrine

The Tu Ka'loh Shrine is a blessing shrine, so there are no challenges. Run ahead and open the treasure chest to get the Barbarian Helm. Then head to the alter to speak with Tu Ka'loh to get the Spirit Orb.
